1. Introduction. |
Review of some concepts. Semiconservative mechanism of replication (Meselson-Stahl experiment). The central dogma of molecular biology. |
2. DNA replication. |
Initiation of replication in bacteria. Multiple origins of replication in eukaryotes and archaea. DNA polymerases and other enzymes involved. Replication mechanism: conductive and delayed chains (Okazaki fragments). Completion of replication. Replication of extra-chromosomal elements (viruses, plasmids and mitochondria).

3. DNA repair. |
Lesions in the DNA. Types of repair systems: Direct inversion, repair by excision, recombination repair, repair of base missmatchings. |
4. DNA transcription. |
Transcription to prokaryotes: initiation, elongation and termination of transcription. RNA polymerase. Promoter sequences. Completion of transcription. Transcription eukaryotes: type of RNA polymerases, transcription factors and promoters of RNA pol. I, II and III, transcription enhancers, transcription termination.

5. RNA processing and maturation. |
Post-transcriptional modifications in eukaryotic 5 'capping, splicing. Spliceosome. Alternative splicing. Trans-splicing. RNA editing. mRNA stability. Processing of rRNA and tRNA.

6. Translation of mRNA into protein. |
Phases of translation: initiation, elongation and termination. The ribosomes. Factors of initiation, elongation and termination. tRNA and aminoacyl-tRNA synthetases. The genetic code. Chaperones and the folding of proteins. Peptide signal and transport of proteins in the endoplasmic reticulum. Post-transductional modifications. Ubiquitin and protein degradation.

7. Regulació de l’expressió gènica. |
Mechanisms of regulation of transcription. Regulation of the operon lac and operated trp in E. coli. Regulation by cascades of sigma factors. Regulation of gene expression in eukaryotes. Epigenetics Regulation by RNAs (riboswitchs, miRNA, siRNA). |
